目的 探讨白色念珠菌蛋白酶活动与其毒力关系。方法的采用牛奶平地检测54株白色念珠菌蛋白酶的分泌能力,然后选择6侏菌分别经摇瓶培养测定其蛋白酶活力,并以静脉内注射方式感染小鼠进行毒力试验,以小鼠平均生存时间来评价菌株毒力。结果 54株白色念珠菌全部能分泌蛋白酶,检出率为100%。动物试验表明蛋白酶活力愈高的菌株,相应小鼠平均生存时间愈短;蛋白酶活力与菌株毒力直接正相关(γ=0.934,P<0.01) 相似文献

发展了一套测定棉铃虫Helicoverpaarmigera (H櫣bner)对植物挥发物的行为反应的生测系统。这套系统由嗅觉仪、风洞和风洞室组成。用这套系统测定了棉铃虫对萎蔫后的枫杨Pterocaryastenoptera叶、加拿大杨Populuscanadensis叶、香椿Toonasisnensis叶、新鲜的芹菜与胡萝卜花挥发物以及苯乙醛的行为反应。结果显示 ,萎蔫的枫杨叶、加拿大杨叶、香椿叶的挥发物均对棉铃虫处女雌蛾表现出显著的引诱作用 ,而对已交配过的雌蛾和雄蛾无明显的引诱作用 ;新鲜的芹菜与胡萝卜花挥发物对棉铃虫两性性成虫均不表现出显著的引诱作用 ;苯乙醛对棉铃虫处女雌、雄蛾有较强的引诱作用。讨论了杨树枝把引诱棉铃虫的原因。 相似文献

Insect-based BioFETs (biologically sensitive field-effect transistors) with improved signal characteristics have been developed. These BioFETs require a specifically adapted signal interfacing between a FET as signal transducer and an intact insect antenna as biocomponent. Therefore, different field-effect transistors have been fabricated in order to study the signal transfer at the bioelectronic interface. As relevant features of the BioFET, its current-voltage characteristics, the transconductance and the signal-to-noise ratio have been investigated as affected by the choice of gate insulator materials and gate dimensions (width-to-length ratio, thickness of the dielectric layers). The performance of the improved FET arrangement in the isolated-antenna BioFET was validated by employing dilution series of the plant odour component Z-3-hexen-1-ol. 相似文献

Cluster of differentiation antigen 4 (CD4), the T lymphocyte antigen receptor component and human immunodeficiency virus coreceptor, is down-modulated when cells are activated by antigen or phorbol esters. During down-modulation CD4 dissociates from p56(lck), undergoes endocytosis through clathrin-coated pits, and is then sorted in early endosomes to late endocytic organelles where it is degraded. Previous studies have suggested that phosphorylation and a dileucine sequence are required for down-modulation. Using transfected HeLa cells, in which CD4 endocytosis can be studied in the absence of p56(lck), we show that the dileucine sequence in the cytoplasmic domain is essential for clathrin-mediated CD4 endocytosis. However, this sequence is only functional as an endocytosis signal when neighboring serine residues are phosphorylated. Phosphoserine is required for rapid endocytosis because CD4 molecules in which the cytoplasmic domain serine residues are substituted with glutamic acid residues are not internalized efficiently. Using surface plasmon resonance, we show that CD4 peptides containing the dileucine sequence bind weakly to clathrin adaptor protein complexes 2 and 1. The affinity of this interaction is increased 350- to 700-fold when the peptides also contain phosphoserine residues. 相似文献

Ultraviolet (UV) radiation at four wavelengths (305, 320, 340 and 380 nm) and photosynthetically active radiation (PAR) were
measured from May 1994 to October 1999 using Biospherical UV radiometers. A surface reference sensor located on the roof of
the Marine Station at Helgoland recorded values every 5 min, and an equivalent profiling underwater sensor was used for measurements
in the sea at approximately monthly intervals. The ratio of 305-nm radiation to PAR varied seasonally, with a 14-fold increase
from winter to summer. A much weaker seasonal trend (ca. 1.5-fold) was apparent in the ratio of 320-nm radiation to PAR, but
there was no seasonal trend in the ratios of 340- or 380-nm radiation to PAR. The year-to-year variations in 305-nm radiation
were also much greater relative to PAR than for the other UV wavelengths, but there was no evidence of a change in the 305 nm:PAR
ratio over the study period. The ratios of both 305- and 320-nm radiation to PAR increased from dawn to midday, but those
of 340- and 380-nm radiation were almost constant through the day, except shortly before sunrise and after sunset when the
proportions of 340- and 380-nm radiation increased. Underwater measurements of PAR and UV suggest that the 1% depth for 305-nm
radiation was little more than 1 m, but this estimate is valid only for summer and autumn because, in other seasons, few reliable
readings for 305-nm radiation could be obtained underwater, and no attenuation coefficient could be calculated. The 1% depths
recorded for the other UV wavelengths in the middle 6 months of the year were 2.0 m for 320 nm, 2.6 m for 340 nm and 4.6 m
for 380 nm, compared with 12 m for PAR, but the attenuation of all wavebands increased sharply in October and remained higher
until March. An analysis of the influence of sun angle, total column ozone concentration, the proportion of skylight, and
cloud cover on the ratio of UV wavelengths to PAR in surface irradiance demonstrated that solar angle has a greater influence
than ozone concentration on the irradiance at 305 nm, and that the typical occurrence of ozone "holes" in spring may not result
in higher UV-B irradiances than occur under higher ozone concentrations in summer. The implications of the data for attempts
to model the biological effects of natural UV radiation on marine organisms are considered.

Summary The predation on spiders in a forest ecosystem by a colony of red wood ants, Formica polyctena, was estimated using a barrier to isolate the colony. Of the ants' total prey, 4.6% were spiders. In order to estimate the effect of F. polyctena within their hunting area on the spider population, the spiders' population density was studied inside and outside the hunting area. Samples of the forest floor were taken, spider webs were counted and pitfall traps were used. Genetic variation of a codominantly inherited pancreas protease, designated CTRA-1, was discovered in the house mouse by isoelectric focusing in polyacrylamide gels. Phenotype CTRA-1A was found in MOLH/Fre and in the majority of common laboratory mouse strains. Phenotype CTRA-1B was found in PWD/Ph. It was characterized by the absence of a corresponding protease band. A third phenotype, CTRA-1C, was observed in IS/Cam and a fourth phenotype, CTRA-1D, was detected in SEG/1. CTRA-1 was found only in the pancreas and may represent the A form of chymotrypsin. The enzyme was shown to be controlled by the presumed structural locus Ctra-1 located on chromosome 8. From two backcross series, including a total of 274 animals, the gene order (Es-1, Es-9)-3.9 +/- 1.7%-Got-2-3.9 +/- 1.7%-(Es-2, Es-7, Es-23)-0.7 +/- 0.5%- Ctra-1-6.3 +/- 2.2%-Prt-2 was established. 相似文献

The principles underlying the assembly of intranuclear compartments are only beginning to be understood. The karyosome is an organelle typical of oocyte nuclei. It represents the tightly packed oocyte chromosomes, arrested at the diplotene of meiotic prophase. It has been known from several insect orders that a prominent capsule of unknown materials is built around the karyosome in the course of previtellogenesis and vitellogenesis. Here we show that F-actin, detected by dye-coupled phalloidin, is a major molecular component of the karyosome capsule in Neuroptera. We investigated capsule formation in six species belonging to the family Chrysopidae. Though F-actin was present in the capsules of all six species there were striking interspecific differences in the morphological array of actin filaments and the developmental dynamics of actin deposition in the capsule. The potential biological significance of the karyosome capsule is discussed with respect to the presence of extrachromosomal rDNA in neuropteran oocytes and the molecular functions known from F-actin. Our results corroborate the still controversial hypothesis of a role for actin as a nuclear protein. 相似文献

Insulin-like growth factor (IGF-I) signaling has been implicated to play an important role in regulation of cardiac growth, hypertrophy, and contractile function and has been linked to the development of age-related congestive heart failure. Here, we address the question to what extent cardiomyocyte-specific IGF-I signaling is essential for maintenance of the structural and functional integrity of the adult murine heart. To investigate the effects of IGF-I signaling in the adult heart without confounding effects due to IGF-I overexpression or adaptation during embryonic and early postnatal development, we inactivated the IGF-I receptor (IGF-IR) by a 4-hydroxytamoxifen-inducible Cre recombinase in adult cardiac myocytes. Efficient inactivation of the IGF-IR (iCMIGF-IRKO) as assessed by Western analysis and real-time PCR went along with reduced IGF-I-dependent Akt and GSK3β phosphorylation. Functional analysis by conductance manometry and MRI revealed no functional alterations in young adult iCMIGF-IRKO mice (age 3 mo). However, when induced in aging mice (11 mo) diastolic cardiac function was depressed. To address the question whether insulin signaling might compensate for the defective IGF-IR signaling, we inactivated β-cells by streptozotocin. However, the diabetes-associated functional depression was similar in control and iCMIGF-IRKO mice. Similarly, analysis of the cardiac gene expression profile on 44K microarrays did not reveal activation of overt adaptive processes. Endogenous IGF-IR signaling is required for conservation of cardiac function of the aging heart, but not for the integrity of cardiac structure and function of young hearts. 相似文献

All functional RNAs are generated from precursor molecules by a plethora of processing steps. The generation of mature RNA molecules by processing is an important layer of gene expression regulation catalysed by ribonucleases. Here, we analysed 5S rRNA processing in the halophilic Archaeon Haloferax volcanii. Earlier experiments showed that the 5S rRNA is cleaved at its 5' end by the endonuclease tRNase Z. Interestingly, a tRNA-like structure was identified upstream of the 5S rRNA that might be used as a processing signal. Here, we show that this tRNA-like element is indeed recognised as a processing signal by tRNase Z. Substrates containing mutations in the tRNA-like sequence are no longer processed, whereas a substrate containing a deletion in the 5S rRNA sequence is still cleaved. Therefore, an intact 5S rRNA structure is not required for processing. Further, we used bioinformatics analyses to identify additional sequences in Haloferax containing tRNA-like structures. This search resulted in the identification of all tRNAs, the tRNA-like structure upstream of the 5S RNA and 47 new tRNA-like structural elements. However, the in vitro processing of selected examples showed no cleavage of these newly identified elements. Thus, tRNA-like elements are not a general processing signal in Haloferax. 相似文献
